ABSTRACT

Objective:To explore the mechanism of the treatment of diabetic feet with thunberg fritillary and honeysuckle based on pharmacological and animal experiments.Methods:The drug ingredients of the drug pair of thunberg fritillary and honeysuckle were obtained from TCMSP, and the targets of the drug pair were obtained from the Swiss target prediction. The targets of the disease target of diabetic foot were screened. The cross target was obtained using Venny 2.1. A protein-protein interaction(PPI) network was constructed by STRING and Cytoscape software. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were carried out by Metascape. The ingredient-target-pathway network was established by Cytoscape software. The diabetic foot rat model was established, and the effects of the treatment and the detection of serum screening targets were observed.Results:The results show that 38 active ingredients in the drug pair acted on 339 disease targets to treat diabetic feet through multiple pathways. The core ingredients include luteolin, pelargonidinin, ziebeimine, peiminine, adenosine, and TP53, EP300, HSP90AA1, CTNNB1, and Akt1 are the critical targets. The results of the genetic function analysis show that the biological process is mainly involved in the combination of cellular transcription factors, the reaction of cells to nitrogen compounds, and hormone reactions. The main components of the cell component mainly involve the film raft, the membrane raft, the small space and so on. The molecular function mainly involves protein kinase activity, phosphate kinase activity, kinase activity and so on. The results of the KEGG enrichment analysis indicate the signaling pathways mainly include PI3K/AKT, HIF-1, EGFR, and MAPK. The local repair effect of the drug pair on the diabetic foot rat model is significant, and the expression of the serum TP53, EP300, HSP90AA1, CTNNB1, Akt1 is significantly increased.Conclusions:The mechanism of the treatment of diabetes foot with thunberg fritillary and honeysuckle is mainly by regulating targets (TP53, EP300, HSP90AA1, CTNNB1 and Akt1), signal pathways (PI3K/AKT, HIF-1, EGFR and MAPK), and biological processes such as enzymatic activity and anti-inflammatory.

ABSTRACT

Objective@# To investigate the inhibitory effect of honeysuckle on Streptococcus mutans UA159 in vitro.@*Methods@# We used a double-dilution method to measure the minimum inhibitory concentration (MIC) of honeysuckle against Streptococcus mutans UA159. Lonicerae lonicerae powder was dissolved in the solvent DMSO, different concentrations of liquid medicine were prepared, and bacterial liquid was added. The solution control group and bacterial liquid control group were set at the same time. The growth and acid production of UA159 were determined using antibacterial experiments. A growth curve and acid production curves were drawn, and the adhesion rate and adhesion inhibition rate were calculated. The effect of honeysuckle on the formation of Streptococcus mutans UA159 was determined by crystal violet quantification, and a microscope and a scanning electron microscope were used to observe biofilm formation and structural changes.@* Results @# The MIC of honeysuckle against Streptococcus mutans UA159 was 12.5 mg/mL. The bacteriostatic experiments showed a difference in the growth, acid production and adhesion of UA159 after honeysuckle treatment (P<0.05) compared with the controls, and the inhibitory effect increased as the drug liquid concentration increased. Crystal violet quantification showed a significant difference in biofilm formation between the pharmaceutical liquid group and the control group (P<0.05). Meanwhile, the forward microscope showed a significant decrease in biofilm formation. Under SEM, the number of bacteria decreased significantly at 0, 6 and 12 h after honeysuckle addition. @*Conclusion @# Honeysuckle inhibits the growth and acid production of UA159 and inhibits adhesion and the formation of biofilms.

ABSTRACT

Objective: To explore the anaphylaxis effect and anaphylaxis substances of honeysuckle. Methods: Rat peritoneal mast cells (PMC) were separated and purified, the cells were incubated with compound 48/80 (0.02 g/L), physiological saline and honeysuckle extract (120 g/L) at 37 °C for 0, 15, 30, 45 and 60 min. Degranulation were observed by optical microscope and transmission electron microscope. Annexin V positive cell rate was detected by flow cytometry to reflect the degranulation rate of PMC. SD rats were supplied with honeysuckle extract by intravenous injection at a dose of 2.25 g/L. After administration, different parameters were analyzed, including the symptoms, histamine (HIS) and tryptase (MCT) levels, which were determined to explore the effect of anaphylaxis. Regression analysis was used to calculate the relationships between the peaks and the pharmacological effects to explore potentially anaphylactoid components. Results: The percentage of Annxin V positive cells and the degranulation ratio were markedly elevated in PMC treated with honeysuckle extract for more than 15 min (P < 0.05). HIS and MCT level were significantly elevated after injection of honeysuckle extract for more than 15 min. Morphology of PMC and systemic symptoms were also changed compared with the controlled group (P < 0.05). Regression analysis was used to calculate the relationship between peaks and pharmacological effects, and to determine peaks 7, 10 and 13 as possible anaphylactoid ingredients. Conclusion: This study established a prospective method to clarify the anaphylactoid components of honeysuckle extract, which would provide guidance for screening anaphylactoid components in traditional Chinese medicine injections containing honeysuckle in the prescription.

ABSTRACT

OBJECTIVE: To develop a gas chromatography (GC) method for determination of chlorfenapyr in flower buds of Lonicera japonica Thunb, and study the degradation dynamics of chlorfenapyr in honeysuckle collected through field experiment, thus providing theoretical references for the application of chlorfenapyr in practical production of honeysuckle. METHODS: Honeysuckle samples were extracted with acetone using a vortex mixer and then cleaned up by passing through a neutral alumina chromatography column, followed by detection by gas chromatography with electronic capture detector (GC-ECD). External standard method was used for quantification. RESULTS: At fortified levels of 0.02-0.5 mg•kg-1, the average recoveries of chlorfenapyr in honeysuckle were from 83.9% to 97.6%, with RSDs of 0.6%-9.2%. The limit of detection was estimated to be 7.3 μg•kg-1. The degradation dynamics of chlorfenapyr in honeysuckle conformed to first-order dynamic equation. The degradation half-life of chlorfenapyr was 2.3 d. CONCLUSION: The method developed in this experiment is simple, rapid, reliable and in line with the requirements of pesticide residue analysis. The degradation rate of chlorfenapyr in honeysuckle is relatively rapid. Chlorfenapyr belongs to pesticides which are liable to degradation.

ABSTRACT

Exogenous calcium can enhance the resistance of certain plants to abiotic stress. Research have demonstrated that exogenous calcium could enhances the resistance of honeysuckle under salt stress by promoting the transmission of photosynthetic electrons.The aim of this study was to investigate the effects of exogenous calcium on the contents of Na~+,K~+,Ca~(2+),Mg~(2+)and the expression of photosynthetic related genes Cab and rbc L. In this study,we used ICP-OES to analysis ion contents and used qRT-PCR to analysis the expression patterns of Cab and rbc L. The results showed that CaCl_2 significantly enhanced the K~+-Na~+,Ca~(2+)-Na~+,Mg~(2+)-Na+ratio of honeysuckle treated with 50 and 100 mmol·L~(-1) NaCl. Meanwhile,Cab and rbc L were significantly up-regulated under short-term salt stress,and CaCl_2 promoted this trend. From the two gene expression patterns,rbc L rapidly up-regulated on the first day of stress and then decreased,and was more sensitive to environmental changes. In summary,exogenous calcium could alleviate salt stress and increase plant development by increasing intracellular K~+-Na~+,Ca~(2+)-Na~+,Mg~(2+)-Na+ratio,and the transient overexpression of Cab and rbc L.

ABSTRACT

Objective To evaluate the effect of different gargling liquid on colony number in air during oral ultrasound scaling for patients with periodontitis.Methods Patients with mild (n=54), moderate (n=54), and severe chronic periodontitis (n=54) were selected, then patients with same degree of periodontitis were randomly divided into three groups, with 18 in each group. Before scaling, patients in each group of mild, moderate, and severe periodontitis were given honeysuckle liquid (gargle A), 3% hydrogen peroxide (gargle B), and normal saline (gargle C) respectively. Bacterial culture and identification of air specimens were conducted at 0 and 30 minutes after the beginning of ultrasonic scaling as well as 10, 20 minutes after the end of ultrasonic scaling.Results Variance analysis of three-factor factorial design data showed that degree of periodontitis, types of gargling liquid, and sampling time had interaction (F=2.666, P=0.002). Comparisons of air colonies: At the beginning of scaling, there was no significant difference in colony number of air around patients using different gargling liquid in mild, moderate and severe groups (all P>0.05). Among patients using the same gargling liquid at 30 minutes after the beginning of scaling, colony number of air was the lowest around patients with mild periodontitis and the highest around patients with severe periodontitis, colony numbers of air around patients with gargle A and B were lower than that of patients with gargle C. Comparison of each sampling points showed that colony number of air around the right side of patients' head was highest, differences were all statistically significant (all P<0.05). 20 minutes after the end of scaling, there was no significant difference in colony number among groups compared before scaling (all P>0.05). The main isolated strains in the air were Streptococcus viridans (32.53%), coagulase negative staphylococcus (24.56%), and filamentous fungi (18.48%).Conclusion There are variety of opportunistic pathogens in the air during oral ultrasound scaling, and the number of colonies is positively correlated with the degree of periodontitis. honeysuckle liquid has a good effect on reducing the number of air colonies during and after scaling.

ABSTRACT

Exogenous calcium can enhance the resistance of certain plants to abiotic stress. However,the role of calcium insaltstressed honeysuckle is unclear. The study is aimed to investigate the effects of exogenous calcium on the biomass,chlorophyll content,gas exchange parameters and chlorophyll fluorescence of honeysuckle under salt stress. The results showed that the calcium-treated honeysuckle had better photochemical properties than the salt-stressed honeysuckle,such as PIABS,PItotal,which represents the overall activity of photosystemⅡ(PSⅡ),and related parameters for characterizing electron transport efficiency φP0,ψE0,φE0,σR,and φR are significantly improved. At the same time,the gas exchange parameters Gs,Ci,Trare also maintained at a high level. In summary,exogenous calcium protects the activity of PSⅡ,promotes the transmission of photosynthetic electrons,and maintains a high Ci,therefore enhances the resistance of honeysuckle under salt stress.

ABSTRACT

BACKGROUND/OBJECTIVES: The honeysuckle berry (HB) contains ascorbic acid and phenolic components, especially anthocyanins, flavonoids, and low-molecular-weight phenolic acids. In order to examine the potential of HB as a hepatoprotective medicinal food, we evaluated the in vitro anti-oxidant and anti-inflammatory activities of Korean HB (HBK) and Chinese HB (HBC). MATERIALS/METHODS: Antioxidant and anti-inflammatory effects of the extracts were examined in HepG2 and RAW 264.7 cells, respectively. The anti-oxidant capacity was determined by DPPH, SOD, CAT, and ARE luciferase activities. The production of nitric oxide (NO) as an inflammatory marker was also evaluated. The Nrf2-mediated mRNA levels of heme oxygenase-1 (HO-1), NAD(P)H dehydrogenase [quinone] 1 (Nqo1), and glutamate-cysteine ligase catalytic subunit (Gclc) were measured. The concentrations of HB extracts used were 3, 10, 30, 100, and 300 µg/mL. RESULTS: The radical scavenging activity of all HB extracts increased in a concentration-dependent manner (P < 0.01 or P < 0.05). SOD (P < 0.05) and CAT (P < 0.01) activities were increased by treatment with 300 µg/mL of each HB extract, when compared to those in the control. NO production was observed in cells pretreated with 100 or 300 µg/mL of HBC and HBK (P < 0.01). Treatment with 300 µg/mL of HBC significantly increased Nqo1 (P < 0.01) and Gclc (P < 0.05) mRNA levels compared to those in the control. Treatment with 300 µg/mL of HBK (P < 0.05) and HBC (P < 0.01) also significantly increased the HO-1 mRNA level compared to that in the control. CONCLUSIONS: Thus, the Korean and Chinese HBs were found to possess favorable in vitro anti-oxidant and anti-inflammatory activities. Nrf2 and its related anti-oxidant genes were associated with both anti-oxidant and anti-inflammatory activities in HB-treated cells. Further studies are needed to confirm these in vivo effects.

ABSTRACT

Objective:To establish the fingerprints of the best anti-MDR-MRSA components in the combination of honeysuckle and forsythia. Methods:The best active anti-MDR-MRSA ingredients of honeysuckle and forsythia were dissolved in 10% methanol water. A Waters Atlantis T3 (250 mm × 4. 6 mm, 5 μm) column was eluted with water and methanol as the mobile phase. The optimal chro-matographic conditions were determined by optimizing the mobile phase and chromatographic conditions. The fingerprints similarity of 10 batches of active anti-MDR-MRSA components in honeysuckle and forsythia was studied. Results:The best active ingredients of the combination of honeysuckle and forsythia showed 7 peaks with promising separation. The similarity of the 10 samples was greater than 0. 9. Conclusion:The fingerprints of the best active anti-MDR-MRSA ingredient in the combination of honeysuckle and forsythia are stable, and can be used for the quality evaluation of the components.

Objective:To analyze the clinical efficacy of compound honeysuckle decoction hot and humid joint fusidic acid cream in the treatment of targeted drugs-induced rash.Methods:80 cases of patients with targeted drugs-induced rash admitted in our hospitalfrom August 2014 to August 2016 were selected and divided into two groups with 40 cases in each group according to the drawing method.The control group was treated by fusidic acid cream,while the observation group was treated by compound honeysuckle decoction hot and humid joint fusidic acid cream,the changes of symptom score,quality of life after treatment,clinical efficacy and incidence of adverse reactions were compared between two groups.Results:After treatment,the symptoms score of observation group ((6.87± 1.25) points) was lower than that of the control group ((10.29± 2.74) points)(P<0.05),the quality of life score ((3.15± 0.57)points) of observation group was lower than that of the control group ((6.42± 1.20) points)(P<0.05).The effective rate of observation group(95.00%) was higher than that of the control group(77.50%)(P<0.05),no statistical difference was found in the incidence of adverse reactions was observed between two groups (P>0.05).Conclusion:Compound honeysuckle decoction hot and humid joint fusidic acid cream was effective in the treatment of targeted drugs-induced rash,it could effectively relieve the clinical symptoms,improve the quality of life.

ABSTRACT

More and more disputes have happened to confront us continuously since the separation of Japanese(wild) honeysuckle flower in Chinese Pharmacopoeia in 2005. The state pharmacopeia committee decided to separate Japanese(wild) honeysuckle flower into two species for japanese(wild) honeysuckle flower, but didn't define their the convincing reasons still did not provide to us as a result that two medicines are not described the differences in natural properties, efficiency and indication, usage and dosage, as well as not given a resolving methodand specific solution. It was known for us that in the history of traditional Chinese medicine(TCM), the phenomenenphenomena of the "one drug from multi-species" and "one species for multi-drug" are very ordinary thingswere ubiquitous. Whether separation of the drug species are separated shall be decided to by clinical efficiency. Through Chinese pharmacopoeia (2015 edition) issue of Chinese Pharmacopoeia the 2015 issue of the Chinese Pharmacopoeia, we still cannot find a scientific solution for the dispute of for Japanese (wild) honeysuckle flower, perhaps because of insufficient reorganization of TCM clinical medication regularities, such as "treatment of different diseases with same drug", " treatment of the same disease with different drugs" and "treatment of the same syndrome with multi-prescriptions", and "one prescription treating multiple syndromes"; lack of in-depth analysis for multi-component TCM compounds and autonomisation of "supermolecular template" for organs and meridians; less attention to the advance of efficacy and safety evaluation technologies for multi-component TCM compounds; impacts from the medication mode of "one ingredient-one composition-one effect"; as well as insufficient research methods for bioequivalent evaluation in preclinic and clinic studies . The dispute for species combination or separation for Japanese(wild) honeysuckle flower was apparently caused by regional economy, drug biological equivalent of efficacy and safety, but arising from clinical principles for systematical syndrome treatment with TCM, or concepts in the treatment of diseases with TCM or western medicines. This paper focused on current studies on Japanese(wild) honeysuckle flower in the combination with TCM clinical medication regularities, such as "treatment of different diseases with same drug", "treatment of the same disease with different drugs" and "treatment of the same syndrome with multi-prescriptions", and "one prescription treating multiple syndromes", expounded the specific pharmacological regularity of "heterogeneous equivalence" of Japanese (wild) honeysuckle flower, and put forward methods for studying bioequivalence of Japanese(wild) honeysuckle flower, in order to solve the combination and separation of Japanese(wild) honeysuckle flower and lay a foundation for promoting the development of Chinese herbal medicine industry.

ABSTRACT

Objective:To establish a rapid and sensitive method for the determination of carbendazim in honeysuckle flowers by LC-MS/MS. Methods:The sample was extracted by acetic ether. The mass spectrometer was operated in the positive ionization elec-trospray ( ESI) mode using selection ion monitoring ( SIM) . The transition m/z 192→160 was used to quantify carbendazim. Results:The method had a satisfactory linearity within the range of 50-5 000 ng·ml-1 for carbendazim (r=0. 999 4), the limit of detection (LOD) was 2. 0 ng·ml-1, and the mean recovery was 93. 2%. Conclusion:The method of LC-MS/MS is sensitive, simple and ac-curate,which is proved to be suitable for the determination of carbendazim in honeysuckle flowers.

ABSTRACT

This study was aimed to investigate the influence on component content indexes,which included chlorogenic acid and luteolin of honeysuckle by the absorption of chemical insecticide thiamethoxam.High-performance liquid chromatography (HPLC) method was used in the analysis of content determination of component content indexes of honeysuckle samples by different concentrations of thiamethoxam.SPSS was used in the statistical analysis.The results showed that compared to samples from the blank control group,the content of chlorogenic acid had the increasing tendency in the honeysuckle groups with different concentrations of thiamethoxam.However,there was no significant difference among different experiment groups.There was no significant difference on luteolin among honeysuckle groups with different concentrations of thiamethoxam and the blank control group.It was concluded that at the recommended dose and high-powered dose of thiamethoxam,there was some influence on the content of chlorogenic acid.However,there was no significant effect on the content of luteoloside.And the quality scores of both contents were within the range defined in the Chinese Pharmacopoeia(2010 edition).

Objective:To establish an HPLC method for the determination of chlorogenic acid in honeysuckle stem and honeysuck-le from different sources in different harvest periods. Methods:A Waters C18 column(250 mm × 4. 6 mm,5 μm)was used. The mo-bile phase consisted of acetonitrile-0. 4% H3 PO4(10:90)with the flow rate of 1. 0 ml·min-1 . The detection wavelength was 327 nm and the column temperature was 30℃. An external standard method was established for the determination of chlorogenic acid in honey-suckle stem and honeysuckle in six different harvest periods from three sources. Results:There was a good linear relationship within the range of 0. 065-1. 300 μg for chlorogenic acid(r=0. 999 8). The content of chlorogenic acid in honeysuckle was the highest in Sep-tember and October. The content of chlorogenic acid in Lonicera acuminata was the highest among the honeysuckle stem from three dif-ferent sources. Conclusion:The content of chlorogenic acid in honeysuckle has a certain relationship with the harvest time,which can provide theoretical basis for the choice of harvest time for honeysuckle stem.

ABSTRACT

This study was aimed to optimize the alcohol precipitation techniques for honeysuckle extract in order to standardize the production of honeysuckle extract of Qingkailing Injection and reduce the differences among batches. The orthogonal test was applied in this study. The content of chlorogenic acid and galuteolin were taken as compre-hensive indicators. Multi-index comprehensive scoring method was used in the data analysis. Three influencing fac-tors, which were the fluid temperature, the stirring speed and the speed of adding alcohol, were optimized in the al-cohol precipitation techniques. The results showed that the optimal alcohol precipitation techniques were when the fluid temperature was 20℃, the stirring speed was 240 rpm and the speed of adding alcohol was one time of the ma-terial per minute. It was concluded that the optimized alcohol precipitation process was stable and feasible.

ABSTRACT

This study was aimed to acquire the dynamic changes of chlorogenic acid and luteoloside of honeysuckle at different collecting periods in order to decide the best harvesting time of honeysuckle in Donghai County, Jiangsu Province. The content determination method used in the detection of chlorogenic acid and luteoloside of honeysuckle was from the 2010 edition of the Chinese Pharmacopoeia. The skills of HPLC fingerprint characteristic features, and the yield of pressed flowers were combined in the evaluation of honeysuckle at the three-green period, two-white pe-riod, great-white period, silver-flower period and the golden-flower period. The results showed that the content of honeysuckle at different blossoming stages had obvious changes in content of chlorogenic acid and luteoloside, as well as the pressed flower quality and yield of the flower. It was concluded that the optimal harvest time of honey-suckle was for the two-white period and the great-white period, which was consistent with the real origin.

OBJECTIVE: To establish a HPLC method for the simultaneous determination of the contents and fingerprint of 7 active constituents (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, isochlorogenic acid B, isochlorogenic acid A, and isochlorogenic acid C) in honeysuckle extract. METHODS: The chromatographic separation was achieved on a AkzoNobel Kromasil C18 column(4.6 mm × 250 mm, 5 μm), the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution with gradient elution at a flow rate of 1.0 mL · min-1, the detection wavelength was set at 326 nm, and the column temperature was 30°C. RESULTS: All the 7 compounds showed good linearity in the ranges of the test concentrations. The RSDs of the precision, stability and reproducibility tests were less than 3%. The average recoveries were in the range of 97.98%-99.29%. CONCLUSION: This method is simple, sensitive, accurate, and can be used for quality control of honeysuckle extract. ABSTRACT

Objective To investigate the effect of intraurethral irrigation with honeysuckle water-decoction on formation of bacterial biofilm on the surface of the catheter. Methods Eighty male patients,who had no history of catheterization and no urinary infection with negative results of urine bacterial culture,were enrolled in the study. All patients were randomly divided into 4 groups with 20 cases in each: group A (patients underwent intraurethral irrigation with honeysuckle water-decoction); group B (intraurethral irrigation with 25% PVP); group C (intraurethral irrigation with normal saline ) and group D (no intraurethral irrigation). The urethral catheters were removed at the 7th day and the bacterial biofilm on the surface of the catheter was examined by scanning electron microscopy (SEM). Results SEM demonstrated uneven- distributed matrixes coated on the surface of the catheter, and bacteria adhered to the matrixes. The average thickness of biofilm in group A, B, C and D was (17.876 ±11.788)μn, (31.090±15. 006)μm,(33.716±11.927)μm, (67.762±23.588)μm, respectively. The average thickness of biofilm in group A, B, C was significantly thinner than that in group D (P<0.01), and the thickness in group A was thinner than that in group B and C (P<0.01), but there was no significance difference between group B and group C (P>0.01). Conclusions Intraurethral irrigation with honeysuckle water-decoction can effectively reduce the formation of bacterial biofilm on the surface of the catheter.

ABSTRACT

Objective To determine the contents of fructose, glucose, sucrose and total saccharide in honeysuckle extracts which were produced by three classical methods, to provide scientific data for producing and controlling of those patent medicines containing honeysuckle extract. Methods Three classical extract methods were used for producing honeysuckle extracts. HPLC-ELSD method was used for determining the contents of fructose, glucose, sucrose, and a spectrophotometric determination method was used for determining the contents of total saccharide. Results The contents of fructose in three honeysuckle extracts were 5.44%, 3.47%, 4.74%, glucose were 4.35%, 2.73%, 4.21%, glucose were 17.41%, 8.47%, 14.47%, and total saccharide were 50.63%, 41.12%, 35.90%, respectively. Conclusion About 36%~51% of total components of honeysuckle extracts were determined except chlorogenic acids, and 19%~27% of components in the extracts were fructose, glucose, sucrose.
